No, you don't want the car running. You need to check resistance from one end to the other end of each wire. You're checking for breaks in the wire, etc. Is that a 3 wire or 2 wire crank position sensor?

3 wire where do i find the other end? i did a visual from the plug to the main harness but couldnt follow it anymore.

It goes to the ECU. If it's a 3 wire, it might be a hall effect sensor in which case it will have power and ground (the third wire is the signal output) so you can check that there is power between those two pins. It's usually 5V for most common types of hall effect sensors, but I'm not really sure what type of sensor is used on the LT1. You should be able to find good troubleshooting information in the factory manual.
